In October 2014, Tanner crab in various condition states were targeted and collected. Targeted crab condition states were visually healthy, visually BCS positive or questionably BCS positive. Crab were held from capture until death (ongoing) and crab were tested regularly for parasite presence using two methods, blood smears and a PCR assay to detect the presence of parasite DNA. Several host blood samples were processed using both histological and biotechnical techniques to determine accuracy and consistency of the assays.\r\n \r\n \r\n \r\n \r\n \r\n \r\n \r\n \r\n \r\n \r\n Crab were marked with two identification tags each and held in 3 tanks at the Ted Stevens Marine Research Institute, Alaska Fisheries Science Center, Juneau, Alaska. Tanks were supplied with water from Auke Bay and maintained at ambient temperature. Crab were checked weekly for mortalities. On a weekly and monthly schedule, hemolymph samples were collected from each crab to test for evidence of the parasite Hematodinium sp. Hemolymph samples were collected using a clean syringe and needle for each crab. Hemolymph was used fresh to make blood smear preparations and/or preserved in ethanol for molecular analysis. Blood smears were dried, stained (see Jensen et al. 2010) and viewed with a compound microscope for the presence of Hematodinium sp. DNA was extracted from ethanol-preserved hemolymph samples and tested for parasite DNA using PCR (see Jensen et al. 2010).\r\n \r\n \r\n \r\n \r\n \r\n \r\n \r\n","source_transform":null,"status":"error"}
